چکیده

For many years the major problem impeding the elucidation of the structure of melanin and the melanosome has been the insolubility of the melanin polymer. To date, alkaline and acid hydrolysis seem to be the only methods available which can completely solubilize the melanosome (1); unfortunately, both methods degrade the constituents of the organelle other than the melanin polymer. The recent emergence of refined techniques for the solubilization of the more insoluble cellular components, such as ribosomes and membranes, justified a reinvestigation of melanosome solubility. This paper reports the successful solubilization of the melanosome, and a partial characterization of its proteins.

Characterization of fish acid proteases by substrate-gel electrophoresis.

Several analytical techniques based upon the use of substrate-polyacrylamide gel electrophoresis were evaluated to achieve characterization of aspartate proteases in fish stomach. Since aspartate proteases of fish are more stable at high pH than mammalian pepsins, the most accurate technique for activity assessment is electrophoresis at neutral pH and revealing of such activity at low pH with h...

Renaturation of telomere-binding proteins after the fractionation by SDS-polyacrylamide gel electrophoresis

A simple method for identification and characterization of telomere-binding proteins is described in this article. After Sodium Dodecyl Sulphate-Polyacrylamide gel electrophoresis (SDS-PAGE), proteins are eluted, renatured and used for retardation analysis with labelled oligonucleotides corresponding to human and plant of telomeric sequences. We show here that this method is efficient to recove...

Vitamin A transport in plasma of the non-mammalian vertebrates: isolation and partial characterization of piscine retinol-binding protein.

Studies were conducted to explore vitamin A transport in the non-mammalian vertebrates, especially Pisces, Amphibia, and Reptilia, and to isolate and partially characterize piscine retinol-binding protein. Retinol-containing proteins in fresh plasma obtained from bullfrogs and a turtle exhibited similar properties to those found in mammalian and chicken plasma: i.e., molecular weight of about 6...

Protein Sequencing from Polyvinylidenedifluoride Membranes: Design and Characterization of a Test Sample (ABRF-9OSEQ) and Evaluation of Results

Technological developments such as two dimensional electrophoresis [11, electroblotting of proteins [2], the introduction of the gas phase sequencer [3], and the introduction of polyvinylidenedifluoride (PVDF) membranes mark major advances of the last decade in protein characterization and isolation. The ability to sequence proteins that have been blotted onto PVDF membranes following their sep...

Isolation and characterization of the gene encoding a novel factor Xa-directed anticoagulant from the yellow fever mosquito, Aedes aegypti.

Mosquito salivary glands secrete a number of proteins that inhibit mammalian hemostasis and facilitate blood feeding. We have isolated the protein product and corresponding cDNA of a gene designated Anticoagulant-factor Xa (AFXa), that encodes the factor Xa (FXa)-directed anticoagulant of the yellow fever mosquito, Aedes aegypti. The protein was purified partially by cation exchange chromatogra...
